its been stated that the stones actively seek each other out, so having them end up in the same place kinda makes sense.
Plus vibranium is still only a powerful metal, it doesn't have any mystical or supernatural properties. It should not allow people to gain powers or talk to the dead in a spirit realm. It only makes sense for the soul stone to have landed in the meteor that hit Wakanda.

I doubt anything but Agents of Shield even references it to be honest.
Nah, that wouldn't apply to this. The Great Mound's origins are at least millions of years ago, and there hasn't been any connections with the other stones in that time in the MCU.
Vibranium doesn't do these things. The Heart-shaped-herb (which grows in vibranium rich soil) is what grants its users enhanced abilities. These attributes you're tying to the soul stone already exist without it in the comics (heart-shaped-herb, vibranium). It could be possible, but who knows; its all just theories at this point. 
Something that I think will be super interesting: In the comics, Wakanda has never been successfully invaded. In Secret Invasion, the skrulls failed bigly:
And in the Infinity story arc, the Black Order also try to invade but fail, while other places like NYC, Atlantis, Doctor Strange's place, and X-Men HQ fall:
We saw Thanos' army invading Wakanda in the Avengers trailer but I think that'll be the one place where the Avengers win and will become a stronghold while the rest of the world falls
