Why I'm just now seeing/getting this level 1 perk in the 4th biome?

You either want max integrity and at least 20% of protection or you want a level 15 weapon with some increased weapon damage and a revive token and its trivial at that point.

On my run I cleared all of B1 with the boss and B2 up until the boss shortcut which has the gate to B3. So by the time I got to the boss in B3 I had over 210% integrity and 30% protection and used a level 15 pistol to take down the boss.

Only because I wanted to take him down fast and the pistol was the only level 15 I could get my hands on easily.
