They saw the blue lightning and determined that it is an effect of a pure Velocity # overdose.
In Earth-1 they just developed their Velocity 9, but over in Earth-2 Wells had developed his own version of "Velocity #" and given it to Jay a long time ago. Jay's the only other person they knew of who overdosed on a Velocity drug.

Wells created the shyt in the first place and knowing what happened to Jay it was obvious at that point. This entire season has revolved around Earth 2, Zoom, and Barry trying to stop him. We know Jay wants to be faster and is dying due to the Velocity serum what more do you need?
