They used multiple lines of evidence, monitoring changing positions of stars in the sky and the curved shadow of Earth during lunar eclipses and changes in location of shadows of other things like a pole in Alexandria.
And then Eratosthenes calculated the circumference of the earth based on his observations of a spherical earth.
